Western blot - Anti-ICAM1 antibody [RP23040065] (AB307993)
Western blot analysis of ICAM-1 using K562 and Raji cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% skim milk for 1 hour at room temperature. ICAM-1 was detected at ~58 kDa using Anti ICAM-1 Antibody (ab307993) at a 1/1000 dilution in 2.5% skim milk at 4°C overnight on a rocking platform. Detection was performed using an HRP-conjugated Goat anti-Rabbit secondary antibody at a 1/5000 dilution and chemiluminescent detection.

Lane 1:
K562 cells at 30 µg
Lane 2:
Raji cells at 30 µg

Western blot analysis of ICAM-1 in whole cell extracts from K562 cells (lane 1), and Raji cells (lane 2) using Anti ICAM-1 Antibody (ab307993) at a dilution of 1 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~58kDa.

Immunocytochemistry/ Immunofluorescence - Anti-ICAM1 antibody [RP23040065] (AB307993)
For immunofluorescence analysis, 90% confluent log phase MDCK cells were fixed with 4% paraformaldehyde for 15 minutes, and blocked with 5% BSA for 1 hour at room temperature. The cells were labeled using ab307993 at 1/500 dilution in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor® 488 Goat anti-Rabbit IgG secondary antibody at a dilution of 1/400 for 30 minutes at room temperature (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI. F-actin (Panel c : red) was stained with Alexa Fluor® 594 phalloidin Panel d is a merged image showing cell junction localization and panel e is a control without primary antibody. The images were captured at 20X magnification.
