ICK also known as Intestinal Cell Kinase is a protein kinase with a mass of approximately 72 kDa. It has an important role in regulating cell cycle progression and cellular signaling. The protein expresses in various tissues including the testis brain and skin where it participates in diverse cellular processes. ICK activity can influence cell division and can impact mitotic regulators further elucidating its role in cellular functions.
Biological function summary

The Intestinal Cell Kinase involves in the process of cilia development and maintenance. It interacts as part of a larger protein complex associated with the ciliary basal body. Within this complex ICK modulates the assembly and disassembly of cilia exerting significant control over ciliary motility and length. Its activity is necessary for maintaining the structure and function of these organelles which play important roles in signal transduction pathways critical to cell movement and sensory functions.

Pathways

ICK proteins sit prominently in the Wnt signaling pathway and the Hedgehog signaling pathway. ICK phosphorylates several substrates including proteins tied to these pathways such as GLI transcription factors involved in Hedgehog signaling. Alterations in ICK activity can lead to disruptions in these pathways and affect processes like cellular differentiation and tissue homeostasis.

ICK mutations associate with human ciliopathies such as Nephronophthisis-related disorders. The dysfunction of ICK within the signaling pathways may also link to cancer development where disruptions can affect cell cycle control. Proteins such as GLI involved in the Hedgehog pathway and impacted by ICK are important in these disorders highlighting the potential of ICK as a therapeutic target.

Regulates the ciliary localization of SHH pathway components as well as the localization of IFT components at ciliary tips (By similarity). Regulates intraflagellar transport (IFT) speed and negatively regulates cilium length in a cAMP and mTORC1 signaling-dependent manner and this regulation requires its kinase activity (By similarity).
