IDH3B also known as isocitrate dehydrogenase 3 (NAD(+)) beta subunit is an enzyme that plays a critical role in the tricarboxylic acid (TCA) cycle. It catalyzes the conversion of isocitrate to alpha-ketoglutarate coupling this with the reduction of NAD+ to NADH. IDH3B has a molecular weight of approximately 41 kDa. Expression of IDH3B is found predominantly in the mitochondria of most eukaryotic cells including in tissues with high energy demands such as muscle and heart.
Biological function summary

IDH3B functions as a part of the IDH3 enzyme complex which consists of alpha beta and gamma subunits. This complex is essential for regulating the TCA cycle affecting cellular energy metabolism. Through its enzymatic activity IDH3B contributes to maintaining proper levels of NADH which in turn influences ATP production and overall cellular respiration.

Pathways

IDH3B serves a significant role in the TCA cycle and is interconnected with the glutamine metabolism pathway. By converting isocitrate to alpha-ketoglutarate it interfaces with proteins such as IDH3A and IDH3G which are subunits of the same enzyme complex. These interactions ensure a well-regulated flow of metabolites necessary for energy production and reductive biosynthesis.

Mutations or dysregulation of IDH3B have been linked to certain types of cancer and neurological disorders. For example some studies have shown alterations in IDH3B expression in gliomas affecting cellular metabolism and growth. Additionally IDH3B interacts with IDH1 and IDH2 which are other isocitrate dehydrogenases known for their mutated forms in various cancers. Plays a structural role to facilitate the assembly and ensure the full activity of the enzyme catalyzing the decarboxylation of isocitrate (ICT) into alpha-ketoglutarate. The heterodimer composed of the alpha (IDH3A) and beta (IDH3B) subunits and the heterodimer composed of the alpha (IDH3A) and gamma (IDH3G) subunits, have considerable basal activity but the full activity of the heterotetramer (containing two subunits of IDH3A, one of IDH3B and one of IDH3G) requires the assembly and cooperative function of both heterodimers.
