Immunoprecipitation - Anti-IDI1 antibody (AB242049)

IDI1 was immunoprecipitated from HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) using ab242049 at 6 μg per reaction. Western blot was performed on the immunoprecipitate using ab242049 at 1 μg/mL.

Supplementary information

IDI1 also known as Isopentenyl-diphosphate delta isomerase 1 plays an important role in isoprenoid biosynthesis. The protein facilitates the conversion of isopentenyl diphosphate (IPP) to dimethylallyl diphosphate (DMAPP) an essential step in the biosynthetic pathway. This enzyme has a molecular mass of approximately 44 kDa. You can find IDI1 expressions mainly in the liver though it is also noted in other tissues including the heart and kidneys.
Biological function summary

IDI1 contributes to the formation of isoprenoids which are essential for numerous cellular functions like cholesterol synthesis and protein prenylation. It principally operates as a monomer and does not typically associate with a larger protein complex during its activity. The enzyme's role in positioning alongside other essential enzymes makes it significant for the transcriptional regulation of certain metabolic processes.

Pathways

IDI1 participates extensively in the mevalonate pathway a fundamental route for cholesterol biosynthesis. It functions under the regulation of proteins like HMG-CoA reductase a rate-limiting enzyme in this pathway. The enzyme also connects to the isoprenoid biosynthesis pathway where it interacts with proteins involved in the synthesis of farnesyl pyrophosphate a precursor in the generation of various biomolecules.

IDI1 links with metabolic conditions such as hypercholesterolemia and Smith-Lemli-Opitz syndrome both affecting cholesterol levels. The interaction with proteins such as DHCR7 involved in the conversion of 7-dehydrocholesterol to cholesterol highlights its implication in these disorders. Dysfunction in IDI1 activity might lead to the disruption of regular metabolic balance further impacting cellular health.
