The protein Immediate Early Response 3 (IER3) also known as IEX1 is an early-response gene product with a mass of approximately 19 kDa. It expresses in various tissues including the skin liver and immune system cells notably in T lymphocytes. IER3 regulates cellular processes such as growth and differentiation by influencing cellular apoptosis and survival mechanisms.
Biological function summary

Immediate Early Response 3 plays a significant role in cellular stress response and immune signaling. IER3 does not form a traditional complex but interacts with intracellular signaling molecules to modulate apoptosis. It also influences the activity of MAPK and NF-kB pathways critical for stress response and inflammation. Through this modulation IER3 directly impacts cell fate under stress conditions.

Pathways

Immediate Early Response 3 is involved in modulating cellular survival and stress response pathways. It interacts with the MAPK signaling pathway which regulates cell growth division and stress response. IER3 influences pathways involving NF-kB a protein complex controlling transcription of DNA and cytokine production. These pathways connect IER3 to other proteins such as Bcl-2 and caspases modifying cellular responses.

Immediate Early Response 3 associates with cancer progression and autoimmune diseases. Its expression levels can influence the development of certain cancers by altering apoptosis and cell proliferation rates. In autoimmune disorders IER3 affects immune responses being linked to proteins like NF-kB which regulates inflammation. Understanding the role of IER3 could aid in developing therapies for conditions like rheumatoid arthritis or certain cancer types.

Target data

May play a role in the ERK signaling pathway by inhibiting the dephosphorylation of ERK by phosphatase PP2A-PPP2R5C holoenzyme. Acts also as an ERK downstream effector mediating survival. As a member of the NUPR1/RELB/IER3 survival pathway, may provide pancreatic ductal adenocarcinoma with remarkable resistance to cell stress, such as starvation or gemcitabine treatment.
