Paraffin-embedded human lung cancer tissue stained for IFITM2 using ab236735 at 1/700 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized tissue using an HRP conjugated SP system.

Biological function summary
The role of IFITM2 extends beyond mechanical blocking as it participates in the immune response by inhibiting the fusion of viral membranes with endosomal membranes. IFITM2 operates together with IFITM1 and IFITM3 forming a complex that boosts its antiviral capacity. The protein contributes to the defense mechanism against a broad range of viral pathogens including influenza and dengue viruses.
Pathways
Research shows that IFITM2 is involved in the interferon signaling pathway which regulates immune response to pathogens. In this pathway IFITM proteins are part of the type I and type II interferon responses enhancing cellular resistance to viral infection. The protein collaborates with others such as STAT1 in maintaining cellular defenses against pathogens.
