IFT122 also known as WDR10 is a subunit of the intraflagellar transport complex B (IFT-B) with a molecular mass of about 141 kDa. It participates in the assembly maintenance and function of cilia which are hair-like structures on cell surfaces that have essential roles in signaling pathways and movement. Expression of IFT122 is observed in many tissues including the kidney liver and nervous system reflecting its importance in diverse biological processes.
Biological function summary

IFT122 plays an integral role in cilia biogenesis and function. As part of the IFT-B complex it helps to transport ciliary proteins to and from the cilia facilitating their proper assembly and function. This transport function is vital for maintaining cilia structure and signaling capabilities which are needed for various cellular responses. Proper functioning of IFT122 ensures that cilia operate effectively in transmitting signals required for cellular communication and development.

Pathways

IFT122 is a central component in the hedgehog signaling pathway and Wnt signaling pathway. These pathways are important for embryonic development and tissue homeostasis. In the hedgehog signaling pathway IFT122 interacts with proteins like Smoothened to regulate gene expression patterns important for developmental processes. The involvement in Wnt signaling also indicates its importance in processes like cell proliferation and differentiation. Both pathways rely on functional cilia highlighting IFT122’s broader impact on cellular signaling dynamics.

Mutations or disruptions in IFT122 are linked to disorders such as cranioectodermal dysplasia (CED) and nephronophthisis. CED affects skeletal formation and tissue integrity implicating the protein's importance in developmental signaling. Nephronophthisis a genetic kidney disease shows how IFT122 dysfunction leads to cyst formation and impairments in kidney function often related to its role in ciliary assembly and signaling. As a component of the IFT complex A (IFT-A), a complex required for retrograde ciliary transport and entry into cilia of G protein-coupled receptors (GPCRs), it is required in ciliogenesis and ciliary protein trafficking (PubMed : 27932497, PubMed : 29220510). Involved in cilia formation during neuronal patterning. Acts as a negative regulator of Shh signaling. Required to recruit TULP3 to primary cilia (By similarity).
