Western blot : Anti-IKKa/b antibody [EPR16628] (ab178870) staining at 1/1000 dilution, shown in green; Mouse anti-GAPDH antibody [6C5] (ab8245) loading control staining at 1/20000 dilution, shown in magenta. In Western blot, ab178870 was shown to bind specifically to IKKa/b. A band was observed at 85 kDa in wild-type HeLa cell lysates with no signal observed at this size in IKKa/b knockout cell line. To generate this image, wild-type and IKKa/b knockout HeLa cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.

Serine kinase that plays an essential role in the NF-kappa-B signaling pathway which is activated by multiple stimuli such as inflammatory cytokines, bacterial or viral products, DNA damages or other cellular stresses (PubMed : 20434986, PubMed : 20797629, PubMed : 21138416, PubMed : 30337470, PubMed : 9346484). Acts as a part of the canonical IKK complex in the conventional pathway of NF-kappa-B activation (PubMed : 9346484). Phosphorylates inhibitors of NF-kappa-B on 2 critical serine residues (PubMed : 20434986, PubMed : 20797629, PubMed : 21138416, PubMed : 9346484). These modifications allow polyubiquitination of the inhibitors and subsequent degradation by the proteasome (PubMed : 20434986, PubMed : 20797629, PubMed : 21138416, PubMed : 9346484). In turn, free NF-kappa-B is translocated into the nucleus and activates the transcription of hundreds of genes involved in immune response, growth control, or protection against apoptosis (PubMed : 20434986, PubMed : 20797629, PubMed : 21138416, PubMed : 9346484).
