Immunocytochemistry/ Immunofluorescence - Anti-IL-10 Antibody [RP23040032] (AB313401)
Immunofluorescent analysis of 70% confluent log phase RAW 264.7 cells treated with LPS (200 µg/mL, 20 hrs) followed by PTI treatment (1X, 4 hrs) fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature labeling IL-10 with ab313401 at 1/100 dilution followed by Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor® 488 at 1/2000 dilution for 45 minutes at room temperature (Panel a : Green). Nuclei (Panel b : Blue) were stained with DAPI. F-actin (Panel c : Red) was stained with Rhodamine Phalloidin at 1/300 dilution. Panel d represents the merged image showing secreted localization. Panel e represents untreated cells with lower expression levels of IL10. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.
