Signals via IL17RA-IL17RC heterodimeric receptor complex, triggering homotypic interaction of IL17RA and IL17RC chains with TRAF3IP2 adapter. This leads to downstream TRAF6-mediated activation of NF-kappa-B and MAPkinase pathways ultimately resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides and matrix metalloproteinases, with potential strong immune inflammation (PubMed:17911633, PubMed:18684971, PubMed:19825828, PubMed:21350122, PubMed:24120361, PubMed:8676080). Plays an important role in connecting T cell-mediated adaptive immunity and acute inflammatory response to destroy extracellular bacteria and fungi. In airway epithelium, mediates neutrophil chemotaxis via induction of CXCL1 and CXCL5 chemokines (By similarity). In secondary lymphoid organs, contributes to germinal center formation by regulating the chemotactic response of B cells to CXCL12 and CXCL13, enhancing retention of B cells within the germinal centers, B cell somatic hypermutation rate and selection toward plasma cells (By similarity). Effector cytokine of a subset of gamma-delta T cells that functions as part of an inflammatory circuit downstream IL1B, TLR2 and IL23A-IL12B to promote neutrophil recruitment for efficient bacterial clearance (By similarity). Immunocytochemistry/Immunofluorescence analysis of A-20 (mouse reticulum sarcoma cell line) cells labelling IL-17A with purified ab79056 at 5 μg/mL. Cells were fixed with 4% paraformaldehyde. A goat anti-rabbit IgG (1/500) was used as the secondary antibody (green). Nuclei counterstained with DAPI (blue).
The higher molecular weight bands seen in the Western blot may represent glycosylated IL-17A.

Immunohistochemical analysis of paraffin-embedded Human Tonsil Tissue using ab79056 at 5 μg/mL. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.
ab79056 staining IL-17A in Dog lymph node tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde, permeabilized with Tween-20 (0.05%) and blocked with 10% serum for 2 hours at 24°C; antigen retrieval was by heat mediation with citrate buffer (0.01M). Samples were incubated with primary antibody (1/100 in PBS +10% serum) for 14 hours at 4°C. A Biotin-conjugated Horse pan-specific anti-IgG polyclonal (1/20) was used as the secondary antibody.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of mouse thymus tissue staining IL-17A with ab79056 at 20μg/ml. Goat anti-rabbit IgG (1/500) was used as the secondary antibody (red).
