This data was developed using ab302922, the same antibody clone in a different buffer formulation.Flow cytometric analysis of 4% paraformaldehyde fixed 90% methanol permeabilized EL4 (mouse lymphoma T lymphocyte) treated with 50ng/ml PMA and 500ng/ml Ionomycin Ca2+ Salt and 500ng/ml Brefeldin A overnight (Right) / Untreated control (Left) cells labelling IL-17A with ab302922 at 1/500 dilution (0.1ug)/ Right and Left (Red) compared with a isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). A Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody.

This data was developed using ab302922, the same antibody clone in a different buffer formulation.Flow cytometric analysis of 2% paraformaldehyde fixed 0.1% Tween-20 permeabilized Mouse thymocytes treated with 50ng/ml PMA and 500ng/ml Ionomycin Ca2+ Salt and 500ng/ml Brefeldin A overnight (Right) / Untreated control (Left) cells labelling IL-17A with ab302922 at 1/500 dilution (0.1ug)/ Right and Left (Red) compared with a isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). A Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody. Cells were surface stained with anti-CD4 conjugated to Alexa Fluor® 647. Then fixed with 2% PFA for 10min followed by intracellularly stained with ab302922.

This data was developed using ab302922, the same antibody clone in a different buffer formulation.Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized EL4 (mouse lymphoma T lymphocyte) cells labelling IL-17A with ab302922 at 1/500 (0.93 ug/ml) dilution, followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 2 ug/mL dilution (Green). Confocal image showing increased cytoplasmic staining in EL4 cells treated with Phorbol-12-myristate-13-acetate (100 ng/mL) and Brefeldin A (500 ng/mL) for overnight is observed. ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 2.5 ug/mL dilution (Red). The Nuclear counterstain was DAPI (Blue). Secondary antibody only control : Secondary antibody is ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 2 ug/mL dilution.

Target data

Effector cytokine of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity (PubMed : 18025225, PubMed : 19144317, PubMed : 26431948). Signals via IL17RA-IL17RC heterodimeric receptor complex, triggering homotypic interaction of IL17RA and IL17RC chains with TRAF3IP2 adapter. This leads to downstream TRAF6-mediated activation of NF-kappa-B and MAPkinase pathways ultimately resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides and matrix metalloproteinases, with potential strong immune inflammation (PubMed : 16200068, PubMed : 17911633, PubMed : 19144317, PubMed : 26431948). Plays an important role in connecting T cell-mediated adaptive immunity and acute inflammatory response to destroy extracellular bacteria and fungi. As a signature effector cytokine of T-helper 17 cells (Th17), primarily induces neutrophil activation and recruitment at infection and inflammatory sites (PubMed : 18025225). In airway epithelium, mediates neutrophil chemotaxis via induction of CXCL1 and CXCL5 chemokines (PubMed : 18025225, PubMed : 27923703). In secondary lymphoid organs, contributes to germinal center formation by regulating the chemotactic response of B cells to CXCL12 and CXCL13, enhancing retention of B cells within the germinal centers, B cell somatic hypermutation rate and selection toward plasma cells (PubMed : 18157131).
