Immunoprecipitation - Anti-IL-18 antibody [EPR19956] (AB207323)
IL-18 was immunoprecipitated from 0.35 mg of mouse spleen lysate with ab207323 at 1/40 dilution. Western blot was performed from the immunoprecipitate using ab207323 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10000 dilution.
Lane 1 : Mouse spleen lysate 10μg (Input).
Lane 2 : ab207323 IP in mouse spleen lysate.
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab207323 in mouse spleen lysate.
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 30 seconds.

Western blot showed AC-YVAD-CMK significantly inhibits the expression of Caspas-1, NLRP-1, IL-1β, and IL-18 in the renal tissues after cecal ligation and puncture. The same blot was stripped and reprobed with actin to confirm equal loading. *p < 0.05 in comparison with the Control group. #p < 0.05 in comparison with the CLP group. N = 6 for each group. The bar represents mean ± s.e.m.
