Western blot - Anti-IL-22RA1 antibody [EPR28682-24] (AB324611)
Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Low expression : TF-1, Jurkat (PMID : 15308104), Caco-2, cerebellum (PMID : 15308104).
Samples are non-boiled as boiling may cause protein aggregation.
In lane 5-12, the lysate was stored at -80℃ prior to Western blotting.
The bands beneath the target band (75 kDa) are likely to be degradation products or non-specific bands.
In lanes 1-4, to minimize protein degradation, tissues were lysed immediately after harvest and used for Western blotting as soon as possible.
The blot of lane 11-12 was developed using a high-sensitivity ECL substrate, allowing for the detection of proteins in the mid-femtogram range.
Lanes 1-10 are incubated with Goat Anti-Rabbit IgG, (H+L), Peroxidase conjugated (ab97051) at 1/20000, lanes 11-12 are incubated with Goat Anti-Rabbit IgG (HRP) with minimal cross-reactivity with human IgG at 1/2000.
In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (ab181602) staining at 1/200000 dilution.

TF-1 (human erythroleukemia erythroblast) whole cell fresh lysate at 48 µg
Lane 4:
Jurkat (human T cell leukemia T lymphocyte from peripheral blood) whole cell fresh lysate at 48 µg
