IL-5RA also known as the interleukin-5 receptor alpha chain or IL-5 receptor is a protein with a critical role in the immune system. It is also referred to as CD125 or CaL40. The protein has an approximate mass of 52 kDa and is expressed mainly on eosinophils and basophils which are types of white blood cells involved in immune responses. The IL-5RA forms a part of the IL-5 receptor complex which also includes a beta chain responsible for signal transduction.
Biological function summary

IL-5RA plays an important role in the growth differentiation and activation of eosinophils. The protein forms part of the IL-5 receptor complex which binds to the cytokine IL-5 initiating a signaling cascade that results in these processes. Through its interaction with IL-5 IL-5RA influences the survival and function of eosinophils cells critical for responses against parasitic infections and in allergic reactions.

Pathways

IL-5RA is fundamental in the JAK-STAT signaling pathway. This pathway is significant as it facilitates the transduction of extracellular cytokine signals into cellular responses impacting the growth and activity of eosinophils. The IL-5 receptor complex is associated with other cytokine receptors through this pathway involving proteins such as JAK2 and STAT5 which play essential roles in the activation of downstream signaling events and cellular responses.

IL-5RA is linked with asthma and eosinophilic esophagitis conditions characterized by an abnormal proliferation and activation of eosinophils. In asthma for example the increased activity of IL-5RA leads to elevated levels of eosinophils contributing to airway inflammation and hyperresponsiveness. IL-5RA also connects with other proteins like IL-5 itself serving as a therapeutic target in treatments aimed at regulating excessive eosinophil activity in these disorders.

Cell surface receptor that plays an important role in the survival, differentiation, and chemotaxis of eosinophils (PubMed : 9378992). Acts by forming a heterodimeric receptor with CSF2RB subunit and subsequently binding to interleukin-5 (PubMed : 1495999, PubMed : 22528658). In unstimulated conditions, interacts constitutively with JAK2. Heterodimeric receptor activation leads to JAK2 stimulation and subsequent activation of the JAK-STAT pathway (PubMed : 9516124).
