Immunohistochemical analysis of paraffin-embedded A : HEK-293T (human epithelial cell line from embryonic kidney) transfected with a human IMPDH1 expression vector containing a Myc-His tag B : HEK-293T (human epithelial cell line from embryonic kidney) transfected with a human IMPDH2 expression vector containing a Myc-His tag C : HEK-293T transfected with empty vector containing a Myc-His tag tissue labeling IMPDH1 with ab325431 at 1/5000 (0.101 µg/ml) dilution, followed by a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection).

Positive staining in A : HEK-293T (human epithelial cell line from embryonic kidney) transfected with a human IMPDH1 expression vector containing a Myc-His tag, and no signal in B : HEK-293T transfected with a human IMPDH2 expression vector containing a Myc-His tag and C : HEK-293T transfected with empty vector containing a Myc-His tag.

The primary antibody was incubated for 30 mins at room temperature.

The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

Counterstained with Hematoxylin.

Secondary antibody only control : Secondary antibody is a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection).

Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins

In Western blot, ab325431 was shown to bind specifically to IMPDH1. Target of interest was observed at 55 kDa in wild-type HAP1 cell lysates (lane 3) with no signal observed at this size in IMPDH1 knockout cell line (lane 4).

Catalyzes the conversion of inosine 5'-phosphate (IMP) to xanthosine 5'-phosphate (XMP), the first committed and rate-limiting step in the de novo synthesis of guanine nucleotides, and therefore plays an important role in the regulation of cell growth. Could also have a single-stranded nucleic acid-binding activity and could play a role in RNA and/or DNA metabolism. It may also have a role in the development of malignancy and the growth progression of some tumors.
