The Influenza A PB1 protein also known as polymerase basic protein 1 forms a core part of the viral polymerase complex. It is a large protein with a molecular weight of approximately 85 kDa. PB1 expresses predominantly in the host cell cytoplasm and nucleus following an Influenza A virus infection. This protein orchestrates the replication and transcription of the viral RNA genome by taking part in the assembly of the viral RNA-dependent RNA polymerase. PB1 interacts with other subunits including PB2 and PA to form this essential polymerase complex.
This viral protein facilitates the copying of viral RNA into progeny genomes and viral mRNA transcripts for protein synthesis. The PB1 component of the polymerase complex incorporates both the template-moving catalytic activities and binding sites. The functional partnerships between PB1 PB2 and PA bring the necessary enzymatic activities for efficient viral gene expression. Its role in the context of the polymerase complex makes it an important factor for the continuation of the viral life cycle ensuring efficient replication of the virus within the host.
PB1 is vital to the viral replication pathway in that it interacts with other polymerase basic proteins like PB2 and related accessory proteins. This pathway oversees the synthesis of viral RNA in both transcriptional and replicative formats. It is closely related to the nuclear export by functioning with the PA and PB2 in the transport of new virus genomes and associated proteins to the host cell cytoplasm eventually leading to the assembly and release of infectious virions.
Influenza A PB1 contributes critically to the pathogenicity and spread of influenza infections in humans and animals. It is notably associated with seasonal flu outbreaks and pandemics. The PB1 protein's interactions with other surface proteins like HA (hemagglutinin) and NA (neuraminidase) influence virulence and transmissibility. Mutations in PB1 can alter the severity and outcome of flu infections making it a target for antiviral drug development and vaccine design aimed at mitigating the impact of influenza-induced respiratory illnesses and complications.

Non-transfected (–) and transfected (+) HEK293T whole cell lysates (30 μg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with Influenza A virus PB1 protein antibody [HL1715] (ab308400) diluted at 1/5000. A HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
