Function

Catalyzes the hydrolysis of the 4-position phosphate of phosphatidylinositol 3,4-bisphosphate (PtdIns(3,4)P2) (PubMed:15716355, PubMed:20463662). Catalyzes also inositol 1,3,4-trisphosphate and inositol 1,4-bisphosphate (By similarity). Antagonizes the PI3K-AKT/PKB signaling pathway by dephosphorylating phosphoinositides and thereby modulating cell cycle progression and cell survival (By similarity) (PubMed:30071275). May protect neurons from excitotoxic cell death by regulating the synaptic localization of cell surface N-methyl-D-aspartate-type glutamate receptors (NMDARs) and NMDAR-mediated excitatory postsynaptic current (By similarity). Isoform 4. Displays no 4-phosphatase activity for PtdIns(3,4)P2, Ins(3,4)P2, or Ins(1,3,4)P3.

Activity summary

INPP4A also known as inositol polyphosphate 4-phosphatase type I is an enzyme that specifically dephosphorylates the D-4 position phosphate of phosphoinositides. It has a molecular mass of about 105 kDa. This enzyme is expressed in several tissues including brain and platelets indicating its involvement in various cellular processes. INPP4A is significant in controlling the intracellular levels of phosphoinositide messengers and plays an important role in signaling pathways.

Biological function summary

INPP4A functions to regulate phosphoinositide signaling which is critical in controlling cell growth apoptosis and other cellular processes. It acts by reducing levels of phosphatidylinositol 34-bisphosphate an important lipid signaling molecule. INPP4A does not appear to form part of a larger complex but operates within a network of other phosphatases and kinases that together maintain lipid signaling homeostasis.

Pathways

INPP4A interacts mainly in the phosphoinositide 3-kinase (PI3K) signaling pathway. This pathway is important for various cellular functions such as metabolism cell growth and survival. INPP4A modifies the activity of related proteins within this pathway like PI3K by reducing phosphatidylinositol 34-bisphosphate levels therefore controlling downstream signaling. It also connects with other phosphoinositide phosphatases like PTEN to finely tune the signaling balance.

Associated diseases and disorders

INPP4A is linked to neurological disorders and cancer. In cancer the dysfunction of INPP4A can lead to abnormal cell growth due to dysregulated PI3K signaling. This enzyme's activity is central in maintaining normal phosphoinositide turnover and its interactions with proteins such as PTEN underline its importance in preventing uncontrolled cell proliferation. For neurological disorders alterations in INPP4A function may affect synapse function and neuronal survival contributing to neurodegenerative processes.
