Flow Cytometry - Anti-Integrin alpha 5 + beta 1 antibody [M200 (Volociximab)] - Rabbit IgG (Chimeric) (AB275977)
PBMC's (A) and U93 cells (B) were stained with unimmunized rabbit IgG antibody (black line) or ab275977 (blue line) at a concentration of 10 μg/ml for 30 mins at RT. After washing, bound antibody was detected using anti-rabbit IgG JK (FITC-conjugate) antibody at 2 μg/ml and cells analyzed on a FACSCanto flow-cytometer.

Immunofluorescence analysis of paraformaldehyde fixed HeLa (human epithelial cell line from cervix adenocarcinoma) cells permeabilized with 0.15% Triton and labeled Integrin alpha 5 + beta 1 with ab275977 at 10 μg/ml for 1h followed by Alexa Fluor®488 secondary antibody (1 μg/ml), showing cytoplasmic and membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom ab275977, DAPI, merged channels and a negative control. The negative control was stained with unimmunized rabbit IgG followed by Alexa Fluor®488 secondary antibody.

Biological function summary
Integrin alpha 5 beta 1 regulates processes like cell adhesion migration and differentiation. It binds to fibronectin in the ECM forming part of the adhesion complexes needed for cellular attachment and survival. This integrin couples with the actin cytoskeleton through talin and vinculin initiating signaling cascades that adjust cellular responses to changes in the microenvironment. Its mechanical linkage to the ECM helps sustain structural integrity affecting tissue homeostasis.
Pathways
Integrin alpha 5 beta 1 participates in the focal adhesion and the MAPK signaling pathway each with significant influence on cell behavior. In focal adhesion it partners with focal adhesion kinase (FAK) an important player in transmitting signals from the ECM to the intracellular environment. The MAPK pathway further involves integrin alpha 5 beta 1 integrating external signals to modulate gene expression and cell cycle progression. These pathways allow the integrin to coordinate cellular processes with the broader tissue context.
