For immunofluorescence analysis, MCF-7 cells were fixed and permeabilized for detection of Integrin alpha 6 using ab308050 followed by detection using an Alexa Fluor 488®-conjugated Goat anti-Rabbit secondary antibody (panel a : green) Nuclei were stained using DAPI (panel b : blue) Actin stained with Alexa Fluor® 594 phalloidin (panel c : red) Panel d is a composite image showing localization of integrin alpha 6 at cell junctions

Western blot - Anti-Integrin alpha 6 antibody [RP23040047] (AB308050)
Western blot analysis of Integrin alpha 6 was performed by loading 20 µg of mouse lung lysate and SW480 cell lysate using a 4-12% Bis-Tris gel, electrophoresis system, and pre-stained protein standard. Proteins were transferred to a PVDF membrane and blocked with 5% skim milk for 1 hour at room temperature. Integrin alpha 6 was detected at ~119 kDa using ab308050 at a 1 : 1000 dilution in 2.5% skim milk at 4°C overnight on a rocking platform. Detection was performed using an HRP-conjugated Goat anti-Rabbit secondary antibody at a 1 : 5000 dilution and chemiluminescent detection was performed (ECL).

Lane 1:
Mouse lung cell lysate at 20 µg

Western blot analysis of Integrin alpha 6 in mouse lung lysate (lane 1) and SW480 whole cell extracts (lane 2) using ab308050 at a dilution of 1 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~119kDa.

For immunofluorescence analysis, 90% confluent log phase MDCK cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0. 25% Triton X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature for detection of Integrin alpha 6 using ab308050 at 1/500 dilution in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor 488® Goat anti-Rabbit IgG secondary antibody at a dilution of 1/400 for 30 minutes at room temperature (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI Panel c is a merged image showing localization at the cell junctions and panel d is a control without primary antibody The images were captured at 20X magnification
