Western blot : Rabbit Monoclonal[EPR9722] to Integrin alpha 9 ab140599 staining at 1/1000 dilution, shown in green; Mouse anti alpha Tubulin (ab7291) loading control staining at 1/20,000 dilution, shown in magenta. A band was observed at 100 kDa in Wild-type A549 cell lysates with no signal observed at this size in ITGA9 knockout A549 cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 100pc LICOR in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW & Goat anti-Mouse 680RD at 1/20,000 dilution.

Product details

ab240116 is the carrier-free version of ab140599.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

Integrin alpha 9 also known as alpha-nine or alpha-9 is a transmembrane protein that is part of the integrin family. It typically forms dimer with beta-1 integrin subunit. This protein has an approximate mass of 150 kDa. Integrin alpha 9 is expressed in various tissues with notable levels in the epithelial cells smooth muscle cells and leukocytes. Its primary mechanical role involves mediating cell adhesion and signaling by interacting with the extracellular matrix (ECM) components and some specific ligands.
Biological function summary

Integrin alpha 9 influences cell migration proliferation and survival through its interactions with ECM proteins like tenascin-C and osteopontin. It does not function alone; it forms part of a complex that includes various signaling molecules and cytoskeletal proteins. These associations facilitate communication between the cell cytoskeleton and the ECM influencing processes like wound healing and inflammation.

Pathways

Integrin alpha 9 plays roles in pathways such as the integrin signaling pathway and the focal adhesion pathway. It interacts closely with molecules like focal adhesion kinase (FAK) and Paxillin which are critical within these pathways. These relationships allow integrin alpha 9 to modulate cellular responses such as spreading migration and cellular structural organization.

Integrin alpha 9 has connections with conditions such as asthma and cancer. In asthma its interaction with ECM proteins contributes to inflammation and airway remodeling. In cancer altered expression and signaling through integrin alpha 9 can influence tumor growth and metastasis. The protein's involvement in disease processes often interacts with other integrins and receptors that regulate inflammation and cellular migration such as certain cytokine receptors.

Target data

Integrin alpha-9/beta-1 (ITGA9 : ITGB1) is a receptor for VCAM1, cytotactin and osteopontin. It recognizes the sequence A-E-I-D-G-I-E-L in cytotactin. ITGA9 : ITGB1 may play a crucial role in SVEP1/polydom-mediated myoblast cell adhesion (By similarity). Integrin ITGA9 : ITGB1 represses PRKCA-mediated L-type voltage-gated channel Ca(2+) influx and ROCK-mediated calcium sensitivity in vascular smooth muscle cells via its interaction with SVEP1, thereby inhibiting vasocontraction (PubMed : 35802072).
