Integrin beta also known as ITGB is a mechanical transducer involved in cell adhesion and signaling. Integrins are heterodimeric proteins consisting of alpha and beta subunits. The human integrin beta subunit has a molecular mass of about 90-110 kDa and it exists in several isoforms such as ITGB1 ITGB2 ITGB3 among others. These integrin beta subunits are widely expressed across various tissues including epithelial cells endothelial cells and immune cells. They mediate interactions between the extracellular matrix and the cytoskeleton inside the cell influencing cellular responses like movement and survival.
Biological function summary

Integrin beta participates actively in mediating cell-extracellular matrix (ECM) interactions as part of the integrin complex. By forming dimers with different alpha subunits integrin beta plays an important role in cellular processes such as differentiation proliferation and apoptosis. The ability of integrin beta subunits to interact with different ECM proteins such as fibronectin and collagen allows them to orchestrate tissue-specific functions. Furthermore integrin beta influences signal transduction pathways leading to activation of intracellular signaling molecules like focal adhesion kinase (FAK) and Src family kinases.

Pathways

Integrin beta significantly impacts cell signaling and communication. It is central to the focal adhesion pathway where it influences the assembly of focal adhesion points that connect cells to the ECM. This connection supports signal cascades that involve Ras-MAPK and PI3K-Akt pathways important for regulating cell proliferation and migration. Integrin beta also interacts with proteins like talin and kindlin enhancing its signaling capabilities and stability at the cell surface thereby affecting cytoskeletal dynamics and cellular motility.

Integrin beta is associated with a range of pathological conditions. Abnormal integrin beta function contributes to cancer progression and metastasis as its altered expression can affect cell adhesion and invasion. Additionally integrin beta dysfunction is implicated in inflammatory diseases like multiple sclerosis where it interacts with proteins such as VCAM-1 to facilitate immune cell infiltration into the central nervous system. Integrin alpha-V/beta-3 (ITGAV : ITGB3) is a receptor for cytotactin, fibronectin, laminin, matrix metalloproteinase-2, osteopontin, osteomodulin, prothrombin, thrombospondin, vitronectin and von Willebrand factor. Integrin alpha-IIb/beta-3 (ITGA2B : ITGB3) is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin.
