INTS10 also known as Integrator Complex Subunit 10 is an integral component of the integrator complex which is involved in the transcriptional process. This protein has a molecular mass of approximately 110 kDa. It expresses widely across various tissues but shows higher expression in certain cell types suggesting a role in specific transcriptional functions. The integrator complex targets RNA polymerase II during the transcription of small nuclear RNAs (snRNAs) which are important for mRNA processing.
Biological function summary

Integrator Complex Subunit 10 contributes to the orchestration of snRNA maturation and termination joining other subunits to form a stable assembly. As part of the integrator complex INTS10 ensures the proper cleavage of snRNA transcripts working alongside partners within the complex. Its functions support RNA processing which is essential for gene expression regulation and cellular response to signals. INTS10 therefore plays a role in maintaining genomic stability through its activities within this complex.

Pathways

INTS10 participates in major transcriptional regulation pathways associated with RNA polymerase II. It influences the RNA processing pathway which is important for snRNA biogenesis. Through its relationship with other integrator subunits such as INTS11 and INTS12 INTS10 facilitates the timely termination of snRNA transcription. This regulation impacts gene expression and can affect pathways like RNA splicing ultimately influencing protein synthesis and cellular functions.

Alterations in INTS10 activity have been connected to neurodevelopmental disorders and certain cancer types. Disruption of the integrator complex often involving INTS11 affects RNA processing leading to transcriptional dysregulation implicated in neurodevelopmental abnormalities. Target data

Component of the integrator complex, a multiprotein complex that terminates RNA polymerase II (Pol II) transcription in the promoter-proximal region of genes (PubMed : 38570683, PubMed : 38823386). The integrator complex provides a quality checkpoint during transcription elongation by driving premature transcription termination of transcripts that are unfavorably configured for transcriptional elongation : the complex terminates transcription by (1) catalyzing dephosphorylation of the C-terminal domain (CTD) of Pol II subunit POLR2A/RPB1 and SUPT5H/SPT5, (2) degrading the exiting nascent RNA transcript via endonuclease activity and (3) promoting the release of Pol II from bound DNA (PubMed : 38570683). The integrator complex is also involved in terminating the synthesis of non-coding Pol II transcripts, such as enhancer RNAs (eRNAs), small nuclear RNAs (snRNAs), telomerase RNAs and long non-coding RNAs (lncRNAs) (PubMed : 16239144, PubMed : 32647223). Within the integrator complex, INTS10 is part of the integrator tail module that acts as a platform for the recruitment of transcription factors at promoters (PubMed : 38823386).
