Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized RAW 264.7 (mouse Abelson murine leukemia virus-induced tumor macrophage) and RAW 264.7 treated with mIFN-gamma (200 ng/mL) and lipopolysaccharide (50 ng/mL) for 24 hr cells labelling IP10 with ab325792 at 1/200 (2.51 µg/ml) dilution, followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 dilution (Green).

ab7291 Anti-alpha Tubulin mouse monoclonal antibody was used to counterstain tubulin at 1/1000 (1 ug/ml) dilution, followed by ab150120 Goat Anti-Mouse IgG H&L (Alexa Fluor® 594) at 1/1000 (2 ug/ml) dilution (Magenta).

Flow cytometric analysis of 4% paraformaldehyde fixed 90% methanol permeabilized 293T cells transfected with a mouse Cxcl10 expression vector containing a myc-His-tag® (Middle) and 293T cells transfected with an empty expression vector containing a myc-His-tag® (Right) cells labelling IP10 with ab325792 at 1/50000 dilution (0.001ug) / Middle and Right compared with a Rabbit monoclonal IgG (ab172730) / Left isotype control.

Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/5000 dilution was used as the secondary antibody.

Cells were co-stained with Myc tag conjugated to Alexa Fluor®647.

Flow cytometric analysis of 4% paraformaldehyde fixed 0.1% Tween-20 permeabilized Mouse bone marrow treated with 100ng/mL M-CSF for 6days (Upper left and right)and Mouse bone marrow treated with 100ng/mL M-CSF for 6days then serum starvation for 2 hours then 50ng/mL IFN beta (Mouse) for 4hours (Lower left and right) cells labelling IP10 with ab325792 at 1/500 dilution (0.1ug) / Right compared with a Rabbit monoclonal IgG (ab172730) / Left isotype control.

Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/5000 dilution was used as the secondary antibody.

Cells were surface stained with anti-CD11b conjugated to BV421. Then fixed with 4% PFA for 10 min followed by intracellularly staining with rabbit IgG or our antibody.

Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/5000 dilution was used as the secondary antibody.

Cells were surface stained with anti-CD86 conjugated to APC/CY7. Then fixed with 4% PFA for 10 min followed by intracellularly staining with rabbit IgG or our antibody.

Pro-inflammatory cytokine that is involved in a wide variety of processes such as chemotaxis, differentiation, and activation of peripheral immune cells, regulation of cell growth, apoptosis and modulation of angiostatic effects (By similarity) (PubMed : 28623423). Plays thereby an important role during viral infections by stimulating the activation and migration of immune cells to the infected sites (PubMed : 18624292, PubMed : 19017990, PubMed : 28468883). Mechanistically, binding of CXCL10 to the CXCR3 receptor activates G protein-mediated signaling and results in downstream activation of phospholipase C-dependent pathway, an increase in intracellular calcium production and actin reorganization. In turn, recruitment of activated Th1 lymphocytes occurs at sites of inflammation (By similarity). Activation of the CXCL10/CXCR3 axis also plays an important role in neurons in response to brain injury for activating microglia, the resident macrophage population of the central nervous system, and directing them to the lesion site. This recruitment is an essential element for neuronal reorganization (PubMed : 15456824).
