IP3R1 or inositol 145-trisphosphate receptor type 1 is a calcium channel that mediates the release of calcium ions from the endoplasmic reticulum into the cytoplasm. It plays critical roles in cellular signaling. Weighing approximately 313 kDa IP3R1 is primarily found in the brain particularly in Purkinje cells of the cerebellar cortex but it also appears in other tissues like heart and liver. This protein belongs to a family of similar receptors IP3R2 and IP3R3 which exhibit different distribution and activity in various tissue types.
Biological function summary

IP3R1 acts as a part of a tetrameric channel complex that senses the presence of inositol trisphosphate (IP3) to initiate calcium release. Its expression and function rely heavily on intracellular calcium regulation which is essential for multiple cellular processes like gene expression metabolism and apoptosis. IP3R1 modulates neuronal function and plasticity by influencing calcium-dependent signal transduction pathways.

Pathways

IP3R1 plays critical roles in the phosphoinositide signaling pathway and the calcium signaling pathway. Target data

Inositol 1,4,5-trisphosphate-gated calcium channel that, upon inositol 1,4,5-trisphosphate binding, mediates calcium release from the endoplasmic reticulum (ER) (PubMed : 10620513, PubMed : 27108797). Undergoes conformational changes upon ligand binding, suggesting structural flexibility that allows the channel to switch from a closed state, capable of interacting with its ligands such as 1,4,5-trisphosphate and calcium, to an open state, capable of transferring calcium ions across the ER membrane (By similarity). Cytoplasmic calcium released from the ER triggers apoptosis by the activation of CAMK2 complex (By similarity). Involved in the regulation of epithelial secretion of electrolytes and fluid through the interaction with AHCYL1 (By similarity).
