IPL-1 also known as STK13 or Aurora C is a serine/threonine protein kinase with a molecular mass of approximately 39 kDa. It expresses mainly in testis and ovarian tissue playing a significant role in cell division. As a member of the Aurora kinase family IPL-1 functions mechanistically in regulating the centrosome during cell cycle progression ensuring the proper segregation of chromosomes during mitosis and meiosis. The protein undergoes phosphorylation to activate its kinase activity which is essential for its role in chromatin modification and assembly of microtubules.
Biological function summary

IPL-1 functions in chromosomal alignment and cytokinesis during cell division. It forms part of the chromosomal passenger complex (CPC) along with INCENP Survivin and Borealin. This complex is essential for correcting errors in chromosome segregation. IPL-1's activity helps maintain genomic stability by ensuring that cells divide correctly. Given its role in the chromosomal passenger complex IPL-1 is critical in monitoring and controlling the accurate distribution of genetic material between daughter cells.

Pathways

IPL-1 is intricately involved in cellular processes like the mitotic spindle checkpoint and the regulation of the mitotic cell cycle. It interacts with other proteins such as Aurora B which shares similar functions in the mitotic pathway. IPL-1 helps coordinate the activities of these pathways and proteins to ensure orderly cell division. Through these pathways IPL-1 influences the balance of cell proliferation and cell death which is important for normal organismal development and function.

Misregulation of IPL-1 is associated with cancers including ovarian and testicular cancer. Aberrant expression or activation of IPL-1 can lead to chromosomal instability contributing to tumorigenesis. Additionally the overexpression of Aurora kinase family members including IPL-1 often co-occurs with Cyclin B1 dysregulation in cancer cells. The connection between IPL-1 and Cyclin B1 highlights potential therapeutic targets for cancer treatment as researchers continue to explore strategies for inhibiting IPL-1 to restore normal cell cycle control in tumor cells.

Serine/threonine-protein kinase component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. The CPC complex has essential functions at the centromere in ensuring correct chromosome alignment and segregation and is required for chromatin-induced microtubule stabilization and spindle assembly. Also plays a role in meiosis and more particularly in spermatogenesis. Has redundant cellular functions with AURKB and can rescue an AURKB knockdown. Like AURKB, AURKC phosphorylates histone H3 at 'Ser-10' and 'Ser-28'. AURKC phosphorylates the CPC complex subunits BIRC5/survivin and INCENP leading to increased AURKC activity. Phosphorylates TACC1, another protein involved in cell division, at 'Ser-228'.
