Supplementary information

IRF2BP1 also known as Interferon Regulatory Factor 2 Binding Protein 1 is a protein involved in regulating gene expression. It has a molecular mass of approximately 64 kDa. This protein plays a role as a transcriptional corepressor meaning it helps repress the activity of certain genes. IRF2BP1 is expressed in various tissues with notable expression in immune cells which suggests its involvement in immune system regulation.
Biological function summary

IRF2BP1 interacts with other proteins to modulate transcriptional repression. It forms part of a complex that includes other co-repressors which collaborate to regulate gene expression tightly. One of its main functions is to inhibit the activity of IRF2 an important transcription factor involved in innate immunity. This inhibition allows for fine-tuning of the immune response and helps maintain immune homeostasis.

Pathways

IRF2BP1 functions within the interferon signaling pathway playing a regulatory role by modulating IRF2 activity. This pathway is important for responding to viral infections and other immune challenges. By interacting with IRF2 IRF2BP1 controls gene expression related to immune response. Additionally IRF2BP1 is involved in pathways related to transcriptional regulation where it may impact the expression of genes under specific conditions.

IRF2BP1 has associations with autoimmune diseases and cancer. Its role in repressing IRF2 suggests that dysregulation could lead to imbalances in the immune response potentially contributing to conditions like systemic lupus erythematosus. Furthermore its activity links to cancer where aberrant expression or function might result in disrupted cell growth and apoptosis. In these contexts IRF1 another immune regulatory protein may interact with IRF2BP1 impacting the outcomes related to such diseases.

Target data

Acts as a transcriptional corepressor in a IRF2-dependent manner; this repression is not mediated by histone deacetylase activities. May act as an E3 ligase towards JDP2, enhancing its polyubiquitination. Represses ATF2-dependent transcriptional activation.
