IRSp53 also known as BAIAP2 (Brain-Specific Angiogenesis Inhibitor 1-Associated Protein 2) is a protein with a molecular mass of approximately 58 kDa. It acts as an important regulator of actin cytoskeleton dynamics facilitating the formation of membrane protrusions like filopodia and lamellipodia. IRSp53 is expressed ubiquitously in various tissues with higher levels observed in the brain. It contains several domains including the IMD (I-BAR) domain which is involved in membrane curvature and actin binding.
IRSp53 participates in the modulation of cell shape and motility. It is a component of the protein complexes that couple membrane dynamics to cytoskeleton remodeling. Through its interaction with proteins like CDC42 and Rac1 IRSp53 guides the rearrangement of actin filaments affecting cell movement and shape. These abilities make it significant for cellular processes requiring changes in cell structure such as neurodevelopment and wound healing.
IRSp53 is central to the regulation of cell motility pathways and signaling processes. It plays an important role in the WAVE (WASP family verprolin-homologous protein) regulatory complex which influences actin polymerization. The protein interacts with Rac1 and N-WASP to mediate pathways leading to the formation of actin-rich structures. This modulation of actin dynamics links IRSp53 to the broader MAPK/ERK signaling pathway impacting cellular proliferation and differentiation.
IRSp53 has been linked to neurological and oncological conditions. Alterations in its expression or function have connections with autism spectrum disorders and glioblastoma. In these disorders IRSp53's interaction with proteins like BAIAP2L1 can influence signal transduction and cytoskeletal organization which may contribute to disease pathology.
