ISL-2 also known as ISLET2 or Islet-2 is a transcription factor with an approximate mass of 39 kDa. It belongs to the LIM/homeobox family and primarily functions by binding DNA to regulate gene transcription. This protein is essential in the central nervous system and is also expressed in the developing spinal cord. In these tissues ISL-2 facilitates the differentiation and maturation of specific neuronal populations by influencing the transcription of downstream target genes.
As part of a complex network of transcription factors ISL-2 plays an important role in neuronal development. It controls genes involved in motor neuron differentiation and axonal guidance. ISL-2 influences the identity and function of neurons participating in the formation of neural circuits. The protein's regulatory capacity ensures correct neuronal patterning impacting the functioning of developing and mature nervous systems.
ISL-2 interacts with signaling pathways related to neuronal specification such as the Hedgehog and Wnt pathways. These pathways are important for the regulation of neural tube development and organization of neuronal structures. In the Wnt pathway ISL-2 interacts with proteins like β-catenin to modulate gene expression important for neuronal identity. Its involvement in these pathways reinforces the protein's function in maintaining nervous system integrity.
ISL-2 has implications in neurodegenerative diseases and spinal muscular atrophy. Mutations or dysregulation of ISL-2 expression can affect motor neuron health leading to progressive loss of motor function. It shows connections with other proteins involved in neurodegeneration such as SMN (survival motor neuron protein) which plays a role in the disease's pathology. Understanding ISL-2's regulation and interactions can offer insights into therapeutic strategies for such disorders.
