Isoleucyl tRNA synthetase also known as IARS or IleRS is an essential enzyme that charges tRNA molecules with isoleucine. This enzyme weighs approximately 126 kDa and is part of the aminoacyl-tRNA synthetase family. By catalyzing the attachment of isoleucine to its corresponding tRNA it ensures the correct translation of mRNA into proteins. IARS is ubiquitously expressed in the cytoplasm of various cell types and tissues playing a critical role in protein synthesis.
IARS plays an important role in maintaining cellular homeostasis by ensuring the fidelity of protein translation. It forms part of a larger complex of aminoacyl-tRNA synthetases known for its multi-enzyme complex formation which assists in proper protein synthesis. Through its function in aminoacylation IARS maintains the balance necessary for cellular activity and division reflecting its importance across multiple biological processes.
IARS has important responsibilities in the translation process which is a fundamental pathway in gene expression. In the context of translation the enzyme contributes to the aminoacyl-tRNA biosynthesis pathway often interacting with other tRNA synthetases to stabilize protein synthesis. Additionally IARS can relate to the mTOR signaling pathway indicating its connection with nutrient-sensing mechanisms and its role in regulating cell growth and proliferation.
Altered IARS function has connections to various neurological disorders such as CARS-related Perrault Syndrome where it may interact with cysteinyl-tRNA synthetase (CARS). Additionally mutations in IARS can relate to liver disorders like Vanishing White Matter (VWM) disease where it is associated with endoplasmic reticulum stress pathways and connected proteins such as eIF2B. Understanding these interactions helps in designing therapeutic strategies targeting the protein for treatment interventions.
