Integrin alpha-V:beta-6 (ITGAV:ITGB6) is a receptor for fibronectin and cytotactin (PubMed:17158881, PubMed:17545607). It recognizes the sequence R-G-D in its ligands (PubMed:17158881, PubMed:17545607). Internalization of integrin alpha-V/beta-6 via clathrin-mediated endocytosis promotes carcinoma cell invasion (PubMed:17158881, PubMed:17545607). ITGAV:ITGB6 acts as a receptor for fibrillin-1 (FBN1) and mediates R-G-D-dependent cell adhesion to FBN1 (PubMed:17158881). Integrin alpha-V:beta-6 (ITGAV:ITGB6) mediates R-G-D-dependent release of transforming growth factor beta-1 (TGF-beta-1) from regulatory Latency-associated peptide (LAP), thereby playing a key role in TGF-beta-1 activation (PubMed:15184403, PubMed:22278742, PubMed:28117447). The protein ITGB6 also known as Integrin beta-6 is a part of the integrin family which are transmembrane receptors. ITGB6 has a molecular mass of approximately 88 kDa. It functions mechanically by binding to extracellular matrix proteins such as fibronectin vitronectin and tenascin. ITGB6 pairs exclusively with the integrin alpha-V subunit to form the integrin alpha-V beta-6 (αvβ6) heterodimer. Its expression mainly occurs in epithelial cells particularly in the skin lungs and gastrointestinal tract.
ITGB6 plays important roles in cell adhesion migration and signaling. It is part of the αv integrin complex which is significant in the regulation of transforming growth factor-beta (TGF-β) activation. This activation influences epithelial-mesenchymal transition (EMT) tissue repair and fibrosis. ITGB6 impacts processes such as inflammation and wound healing due to its expression in epithelial tissues thereby contributing to maintaining tissue integrity and function.
ITGB6 interacts in the TGF-β signaling and epithelial cell migration pathways. TGF-β activation via ITGB6 plays a critical role in controlling immune responses and cellular proliferation. ITGB6 is interconnected with other integrin family members such as ITGAV (Integrin alpha-V) which collaborates in cellular mechanisms involving cell adhesion and signaling processes. These interactions contribute to cellular responses to extracellular signals that influence pathophysiological processes.
ITGB6 has associations with fibrosis and cancer. In fibrotic diseases ITGB6 facilitates excess activation of TGF-β leading to aberrant tissue remodeling in organs such as the lungs and liver. Regarding cancer ITGB6 influences tumor progression and metastasis particularly in epithelial-derived cancers due to its role in EMT and tissue invasion. ITGB6's interactions with other integrins and ECM proteins contribute to tumorigenesis and fibrotic tissue changes making it a therapeutic target in these conditions.
