Activity summary

ITIH2 also known as Inter-alpha-trypsin inhibitor heavy chain H2 is a protein involved in matrix stabilization during inflammation and tissue repair. With a mass of approximately 125 kDa this protein is primarily expressed in the liver and secreted into the bloodstream. It forms part of the inter-alpha-trypsin inhibitor family where it binds to small glycosaminoglycans such as hyaluronan playing a role in the structural integrity of extracellular matrices.

Biological function summary

ITIH2 functions as a stabilizer in extracellular matrices especially during inflammation and tissue repair processes. It is part of a complex known as the serum-derived hyaluronan-associated protein (SHAP) which contributes to the VCAN (versican) network of the extracellular matrix. This complex participates in organizing matrix assembly and modulating cellular responses during tissue injury.

Pathways

In inflammatory and repair mechanisms ITIH2 plays an important role. It engages in pathways involving extracellular matrix degradation and repair as well as inflammatory response signaling. ITIH2 interacts with proteins such as hyaluronan synthase to influence matrix dynamics. This interaction highlights its participation in significant pathways like those regulating tissue turnover and inflammatory responses.

Associated diseases and disorders

ITIH2 has been linked to inflammatory conditions and liver disorders. In the context of liver diseases altered levels of ITIH2 may reflect liver pathology such as cirrhosis or hepatitis.
