Inositol 1,4,5-trisphosphate-gated calcium channel that upon inositol 1,4,5-trisphosphate binding transports calcium from the endoplasmic reticulum lumen to cytoplasm (PubMed:25329695). Exists in two states; a long-lived closed state where the channel is essentially 'parked' with only very rare visits to an open state and that ligands facilitate the transition from the 'parked' state into a 'drive' mode represented by periods of bursting activity (By similarity).

The type 2 inositol 145-trisphosphate receptor (ITPR2) acts as a channel for calcium ions. This receptor is also known as IP3R2 and weighs approximately 313 kDa. It consists of six transmembrane domains. ITPR2 enables the release of calcium from the endoplasmic reticulum into the cytosol. ITPR2 is expressed mostly in the brain heart and testis although its expression can vary in other tissues according to physiological needs.
ITPR2 regulates intracellular calcium signaling. Calcium ions serve as second messengers in a variety of cellular processes. ITPR2 forms a functional tetrameric complex which increases its functional diversity. This complex plays a role in neuronal communication muscle contraction and various cellular signaling pathways like cell growth.
ITPR2 participates actively in the calcium signaling pathway. It closely associates with proteins such as calmodulin that mediate cellular responses. ITPR2 is also central to the phosphoinositide pathway where it links with phospholipase C to initiate downstream effects. These pathways impact diverse physiological functions by modulating intracellular calcium levels.
Abnormal function of ITPR2 links to neurodegenerative diseases such as Alzheimer's disease. It can lead to disrupted calcium homeostasis affecting neuronal viability. ITPR2 also associates with cardiac hypertrophy where calcium imbalance impairs cardiac cell function. Interactions with proteins like amyloid-beta in Alzheimer's contribute to its pathological role. Understanding these interactions could offer targets for therapeutic intervention.

4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilised SH-SY5Y (human neuroblastoma cell line from bone marrow) cells labeling ITPR2 using ab5805 at 2 μg/ml in 0.1% BSAfor 3 hours at RT, followed by a Goat Anti-Rabbit (H&L) AlexaFluor® secondary antibody (Green (a)). The nuclear counter stain is DAPI (Blue (b)). F-actin is stained with AlexaFluor® 555-Rhodamine (Red (c)). Panel D shows the merged image. Panel (e) is a secondary antibody only control.
