Function

Probable protease subunit of the COP9 signalosome complex (CSN), a complex involved in various cellular and developmental processes. The CSN complex is an essential regulator of the ubiquitin (Ubl) conjugation pathway by mediating the deneddylation of the cullin subunits of the SCF-type E3 ligase complexes, leading to decrease the Ubl ligase activity of SCF-type complexes such as SCF, CSA or DDB2. The complex is also involved in phosphorylation of p53/TP53, c-jun/JUN, IkappaBalpha/NFKBIA, ITPK1 and IRF8, possibly via its association with CK2 and PKD kinases. CSN-dependent phosphorylation of TP53 and JUN promotes and protects degradation by the Ubl system, respectively. In the complex, it probably acts as the catalytic center that mediates the cleavage of Nedd8 from cullins. It however has no metalloprotease activity by itself and requires the other subunits of the CSN complex. Interacts directly with a large number of proteins that are regulated by the CSN complex, confirming a key role in the complex. Promotes the proteasomal degradation of BRSK2.

Activity summary

JAB1 also known as c-Jun activation domain-binding protein 1 or COP9 signalosome complex subunit 5 (COPS5) plays a mechanical role as part of the COP9 signalosome. It has a mass of around 37 kDa. JAB1 expresses in various tissues and connects with several proteins for controlling diverse cellular processes. The protein acts as a coactivator for transcription factors like c-Jun playing a part in cellular response to environmental changes.

Biological function summary

The COP9 signalosome of which JAB1 is a part regulates protein degradation by modifying the activity of cullin-RING ubiquitin ligases. This complex plays an important part in processes like cell cycle progression and signal transduction. JAB1 influences cellular responses to stimuli by affecting the degradation of key regulatory proteins making it a component in maintaining protein homeostasis.

Pathways

JAB1 integrates into signaling pathways like the TGF-β signaling pathway and the cell cycle regulation pathway. JAB1 interacts with other proteins including Smad7 in the TGF-β pathway facilitating the degradation of inhibitory Smads and therefore modulating the signal transduction. Through such interactions JAB1 contributes to the regulation of cellular proliferation and differentiation.

Associated diseases and disorders

JAB1 relates to cancer progression and neurodegenerative diseases. Overexpression and misregulation can contribute to tumorigenesis as it influences the stability of proteins involved in cell growth. In the context of neurodegenerative diseases JAB1 interacts with proteins like p27kip1 impacting neuronal survival. Understanding these connections aids in studying their impact on disease development and progression.

    We have systematically measured KD (the equilibrium dissociation constant between the antibody and its antigen), of more than 840 recombinant antibodies to assess not only their individual KD values but also to see the average affinity of antibody.
    Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.
