Endoplasmic reticulum transmembrane protein involved in vesicle-mediated transport, which is required for neutrophil function. Required for vesicle-mediated transport; it is however unclear whether it is involved in early secretory pathway or intracellular protein transport. Acts as a regulator of neutrophil function, probably via its role in vesicle-mediated transport: required for defense against fungal pathogens and for granulocyte colony-stimulating factor (GM-CSF) signaling pathway; possibly by regulating glycosylation and/or targeting of proteins contributing to the viability and migration of neutrophils.

JAGN1 also known as Jagunal homolog 1 is a protein expressed in various tissues including the bone marrow and immune cells. The protein has a mass of approximately 18 kDa. JAGN1 plays a role in the endoplasmic reticulum where it is involved in the process of protein maturation and trafficking. It is a vital component in ensuring proper glycosylation of proteins which is important for their stability and function.
The protein aids in cellular processes linked to immune cell function and development. It forms part of a complex that participates in the secretory pathway influencing how proteins are processed and transported within cells. This involvement directly impacts the immune response as it ensures that immune cells mature correctly and are functionally active.
JAGN1 plays a significant role in the N-linked glycosylation pathway an important process necessary for protein folding and stability. It interacts with other proteins like Dolichyl-Diphosphooligosaccharide-Protein Glycosyltransferase to effectively carry out its glycosylation functions. Another important pathway includes the unfolded protein response pathway which involves proteins like IRE1 and PERK important for maintaining cellular homeostasis under stress conditions.
JAGN1 has been linked to severe congenital neutropenia a disorder affecting the production and function of neutrophils important immune cells. Mutations in the JAGN1 gene have been observed in patients with this condition leading to increased vulnerability to infections. Furthermore research suggests a potential connection with asthma where it may affect immune cell function and inflammatory responses. Through these conditions JAGN1 interacts with proteins like ELANE and CSF3R further elucidating its importance in immune-related diseases.
