JNK1 and JNK3 also known as c-Jun N-terminal kinase 1 and 3 are important members of the mitogen-activated protein kinase (MAPK) family. JNK1 has a mass of approximately 48 kDa while JNK3 is slightly larger around 55 kDa. Both are widely expressed throughout various tissues. However JNK3 is particularly abundant in the nervous system with significant presence within the brain and neuronal tissues.
Biological function summary

These kinases play key roles in regulating several cellular processes including cell growth differentiation and apoptosis. JNK1 and JNK3 function as part of larger signaling complexes facilitating their roles through phosphorylation of specific transcription factors such as c-Jun. These interactions enable cells to respond adaptively to stress stimuli such as ultraviolet radiation and cytokines by modulating gene expression pertinent to these stresses.

Pathways

JNK1 and JNK3 are central to the MAPK signaling cascade. They integrate signals from both the extracellular environment and internal stress signals. Their activity influences pathways like the stress-activated protein kinase (SAPK) cascade and the mitochondrial apoptosis pathway. These kinases also interact with proteins such as p53 further linking them to key mechanisms underlying cellular response and repair processes.

JNK1 and JNK3 have strong associations with neurodegenerative conditions and cancer. Serine/threonine-protein kinase involved in various processes such as cell proliferation, differentiation, migration, transformation and programmed cell death (PubMed : 28943315, PubMed : 9393873). Extracellular stimuli such as pro-inflammatory cytokines or physical stress stimulate the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway. In this cascade, two dual specificity kinases MAP2K4/MKK4 and MAP2K7/MKK7 phosphorylate and activate MAPK8/JNK1. In turn, MAPK8/JNK1 phosphorylates a number of transcription factors, primarily components of AP-1 such as JUN, JDP2 and ATF2 and thus regulates AP-1 transcriptional activity (PubMed : 11602244). Phosphorylates the replication licensing factor CDT1, inhibiting the interaction between CDT1 and the histone H4 acetylase HBO1 to replication origins. Loss of this interaction abrogates the acetylation required for replication initiation. Promotes stressed cell apoptosis by phosphorylating key regulatory factors including p53/TP53 and Yes-associates protein YAP1.
