Supplementary information

K33A also known as Katanin p60 subunit A1 is a microtubule-severing ATPase that plays a significant role in cellular function. This protein has a mass of approximately 60 kDa and is expressed in a variety of tissues with particular abundance in neural tissues. K33A functions by cutting microtubules which are essential components of the cytoskeleton into smaller pieces facilitating cellular processes like mitosis and migration.
Biological function summary

The ability of K33A to sever microtubules affects the dynamic restructuring of the cytoskeleton which is essential for cell division and intracellular transport. It often operates as part of a protein complex with other proteins such as Katanin p80 which regulates its activity. This activity is important for maintaining proper microtubule network organization which influences cellular architecture and function.

Pathways

The severing action K33A performs is essential in the regulation of the microtubule dynamics pathway. This pathway plays a critical role in cellular events like signal transduction where K33A interacts with and modulates the behavior of microtubules. K33A also indirectly connects with proteins like Tau which stabilizes microtubules suggesting its significant involvement in maintaining cellular stability and transport.

Mutations or dysregulation in K33A expression have links to neurological diseases such as spastic paraplegia and neurodegeneration disorders. Neurodegeneration often involves the abnormal behavior of K33A's interacting protein Tau leading to disrupted cellular transport and stability. Understanding these connections emphasizes the importance of K33A in neuronal health and disease pathology.
