Kallistatin also known as PI-4 is a serine proteinase inhibitor with a mass of approximately 52 kDa. You find it mainly in the liver as part of the serpin superfamily. This protein acts mechanically by inhibiting the tissue kallikrein-kinin system which involves proteolytic processes that regulate vascular homeostasis and blood pressure. In addition to the liver kallistatin is expressed in endothelial cells and various human tissues.
Biological function summary

Kallistatin participates in inhibiting angiogenesis and inflammation impacting cellular behavior and responses. As a serpin it functions uniquely in forming stable complexes with target enzymes like tissue kallikrein. Through this mechanism kallistatin contributes to maintaining vascular integrity and preventing excessive vascular growth or inflammation that could lead to pathology.

Inhibits human amidolytic and kininogenase activities of tissue kallikrein. Inhibition is achieved by formation of an equimolar, heat- and SDS-stable complex between the inhibitor and the enzyme, and generation of a small C-terminal fragment of the inhibitor due to cleavage at the reactive site by tissue kallikrein.
