Nuclear receptor coactivator that directly binds nuclear receptors and stimulates the transcriptional activities in a hormone-dependent fashion. Involved in the coactivation of different nuclear receptors, such as for steroids (PGR, GR and ER), retinoids (RXRs), thyroid hormone (TRs) and prostanoids (PPARs). Also involved in coactivation mediated by STAT3, STAT5A, STAT5B and STAT6 transcription factors. Displays histone acetyltransferase activity toward H3 and H4; the relevance of such activity remains however unclear. Plays a central role in creating multisubunit coactivator complexes that act via remodeling of chromatin, and possibly acts by participating in both chromatin remodeling and recruitment of general transcription factors. Required with NCOA2 to control energy balance between white and brown adipose tissues. Required for mediating steroid hormone response. Isoform 2 has a higher thyroid hormone-dependent transactivation activity than isoform 1 and isoform 3.

KAT13A also known as SRC1 or nuclear receptor coactivator 1 (NCOA1) is a coactivator protein with an approximate mass of 160 kDa. This protein aids in the function of nuclear receptors and exerts its mechanical roles by interacting with transcription factors facilitating transcriptional activation. KAT13A is widely expressed in tissues such as the liver skeletal muscle and brain highlighting its ubiquitous role in cellular processes.
KAT13A modulates gene expression by forming part of multi-protein complexes significantly affecting cellular functions like growth and differentiation. The protein serves as a bridge between nuclear receptors and the transcriptional machinery effectively enhancing transcription upon binding. It also plays a role in steroid hormone responses impacting pathways involved in metabolism and inflammation.
KAT13A participates in the steroid hormone receptor signaling pathway and the PPAR signaling pathway. Within these pathways it interacts with proteins such as the estrogen receptor and PPAR-alpha contributing to energy homeostasis and regulating lipid metabolism. These interactions position KAT13A as an important player in integrating hormonal signals with transcriptional response.
KAT13A's dysregulation associates with cancer particularly breast cancer and metabolic disorders like obesity. Its relationship with the estrogen receptor links KAT13A to hormone-dependent cancers affecting cancer cell proliferation. Additionally changes in PPAR-alpha interaction relate to metabolic syndrome influencing lipid metabolism and contributing to the development of obesity-related complications.

Blocking and diluting buffer and concentration: 5% NFDM/TBST
The bands beneath the target band (180 kDa) are likely to be degraded target fragments.
Lysates were freshly made and used immediately to minimize protein degradation.
