Paraffin-embedded human kidney tissue stained for KAT9 / Elp3 using ab236800 at 1/400 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

KAT9 also known as Elp3 is an important catalytic component of the elongator complex which is involved in transcriptional elongation. This protein possesses histone acetyltransferase (HAT) activity enabling it to modify histones by acetylating lysine residues a process important for chromatin remodeling and regulation of gene expression. KAT9 is composed of around 55 kDa and is ubiquitously expressed in various tissues reflecting its essential role in diverse cellular processes.
Biological function summary

KAT9/Elp3 is integral in regulating normal cellular growth and differentiation. It functions as part of the elongator complex which consists of six subunits playing a significant role in tRNA modification and mRNA translation elongation. By supporting the translational machinery Elp3 influences cell cycle progression which is critical for proper cellular functions and organism development.

Pathways

The functions of KAT9 hold relevance in the transcriptional and translational regulation pathways. It contributes significantly to the RNA polymerase II-mediated transcription elongation pathway. In addition there is interaction with proteins such as ELP1 and ELP2 with which it forms complexes facilitating various stages of RNA processing and modification. The activity of these complexes impacts cellular responses under stress conditions regulating gene expression for adaptation.

Alterations in KAT9 expression or function are associated with neurodegenerative diseases and cancers. Defects in the elongator complex involving Elp3 relate to familial dysautonomia a disorder affecting the autonomic nervous system. Additionally KAT9 is connected to colorectal cancer where its dysregulation alters transcriptional dynamics potentially contributing to tumor progression. The interplay between Elp3 and other elongator components like Elp1 highlights its significance in maintaining normal cellular homeostasis and preventing pathological states.

Catalytic tRNA acetyltransferase subunit of the elongator complex which is required for multiple tRNA modifications, including mcm5U (5-methoxycarbonylmethyl uridine), mcm5s2U (5-methoxycarbonylmethyl-2-thiouridine), and ncm5U (5-carbamoylmethyl uridine) (PubMed : 29415125). In the elongator complex, acts as a tRNA uridine(34) acetyltransferase by mediating formation of carboxymethyluridine in the wobble base at position 34 in tRNAs (By similarity). May also act as a protein lysine acetyltransferase by mediating acetylation of target proteins; such activity is however unclear in vivo and recent evidences suggest that ELP3 primarily acts as a tRNA acetyltransferase (PubMed : 29415125). Not a major acetyltransferase of tubulin (PubMed : 22153374). Involved in neurogenesis : regulates the migration and branching of projection neurons in the developing cerebral cortex, through a process depending on alpha-tubulin acetylation (PubMed : 19185337). Required for acetylation of GJA1 in the developing cerebral cortex (By similarity).
