Supplementary information

KCNA3 also known as Kv1.3 is a voltage-gated potassium channel that plays a part in controlling membrane potential in cells. It consists of a protein with a mass of approximately 57 kDa. KCNA3 is mainly expressed in the immune system particularly in T lymphocytes macrophages and microglia. Its expression also extends to the central nervous system where it helps regulate neuronal excitability.
Biological function summary

This potassium channel contributes to the activation and proliferation of T cells. It typically forms a complex with other ion channels to coordinate cellular responses to stimuli. KCNA3 regulates calcium signaling and helps balance the cell's internal environment by facilitating potassium efflux providing support for various immune functions.

Pathways

KCNA3 is involved in both the immune response and intracellular signaling pathways. It plays roles in pathways like the calcium signaling pathway and the MAPK signal transduction cascade. Through these pathways it often interacts with other proteins such as calcium release-activated calcium (CRAC) channels to maintain cellular homeostasis and function.

KCNA3 has associations with autoimmune conditions like multiple sclerosis and rheumatoid arthritis. Dysfunction in KCNA3's regulation of T cell activity can lead to abnormal immune responses. It's linked with the protein CTLA-4 which is another immune checkpoint molecule involved in autoimmune pathologies. This makes KCNA3 a potential target for therapy in related autoimmune diseases.

Target data

Isoform 1. Mediates the voltage-dependent potassium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a potassium-selective channel through which potassium ions may pass in accordance with their electrochemical gradient.. Isoform 2. Mediates the voltage-dependent potassium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a potassium-selective channel through which potassium ions may pass in accordance with their electrochemical gradient.. Isoform 3. Lacks voltage-gated potassium channel activity.
