Immunohistochemical analysis of paraffin-embedded (A) HEK-293T cells t tissue labeling KCNQ3 with ab302782 at 1/8000 (0.066 ug/ml) followed by a LeicaDS9800 (Bond™ Polymer Refine Detection) was used at Ready to use dilution. Positive staining on (A) HEK-293T cells transfected with a mouse KCNQ3 expression vector containing a his tag. No staining on (B) HEK-293T cells transfected with a mouse KCNQ2 expression vector containing a his tag and (C) HEK-293T cells transfected with empty vector containing a his tag. The section was incubated with ab302782 for 30 mins at room temperature.The immunostaining was performed on a Leica Biosystems BOND® RX instrument Counterstained with Hematoxylin. Secondary antibody only control : Secondary antibody is LeicaDS9800 (Bond™ Polymer Refine Detection) was used at Ready to use dilution. Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins

Target data

Pore-forming subunit of the voltage-gated potassium (Kv) M-channel which is responsible for the M-current, a key controller of neuronal excitability. M-channel is composed of pore-forming subunits KCNQ2 and KCNQ3 assembled as heterotetramers (By similarity). The native M-current has a slowly activating and deactivating potassium conductance which plays a critical role in determining the subthreshold electrical excitability of neurons as well as the responsiveness to synaptic inputs. M-channel is selectively permeable in vitro to other cations besides potassium, in decreasing order of affinity K(+) > Rb(+) > Cs(+) > Na(+). M-channel association with SLC5A3/SMIT1 alters channel ion selectivity, increasing Na(+) and Cs(+) permeation relative to K(+). Suppressed by activation of M1 muscarinic acetylcholine receptors. KCNQ3 also associates with KCNQ5 to form a functional channel in vitro and may also contribute to the M-current in brain (By similarity).
