Immunofluorescent analysis of SK-OV-3 cells fixed using 4% formaldehyde (reconstituted in 1X PBS) for 10 min at room temperature and permeabilized using 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling JMJD2B with ab313553 at 1/100 dilution followed by Goat anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 at 1/2000 dilution. Panel a) shows representative cells that were stained for detection and localization of JMJD2B protein (green), Panel b) is stained for nuclei (blue) using DAPI. Panel c) represents cytoskeletal F-actin staining using Rhodamine Phalloidin at 1/300 dilution. Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of JMJD2B. Panel e) represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

Chromatin Immunoprecipitation (ChIP) assay of endogenous KDM4B protein using ab313553/ ChIP was performed using 5 µg ab313553 on sheared chromatin from 2 million PANC1 cells using the MAGnify ChIP System kit. Normal Rabbit IgG was used as a negative IP control. The purified DNA was analyzed by qPCR using primers binding to JUNB and ZEB1 transcriptional start site, PUMA intron 1 and SAT2 satellite repeats. Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method.

Target data

Histone demethylase that specifically demethylates 'Lys-9' of histone H3, thereby playing a role in histone code. Does not demethylate histone H3 'Lys-4', H3 'Lys-27', H3 'Lys-36' nor H4 'Lys-20'. Only able to demethylate trimethylated H3 'Lys-9', with a weaker activity than KDM4A, KDM4C and KDM4D. Demethylation of Lys residue generates formaldehyde and succinate (PubMed : 16603238, PubMed : 28262558). Plays a critical role in the development of the central nervous system (CNS).
