Immunofluorescent analysis of LNCaP cells fixed using 4% formaldehyde (reconstituted in 1X PBS) for 10 min at room temperature and permeabilized using 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling JIK with ab313545 at 1/100 dilution and labeled with Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor 488 conjugate (Product # A27034, 1/2000). Nuclei (blue) were stained using DAPI , and Rhodamine Phalloidin at 1/300 dilution was used for cytoskeletal F-actin (red) staining. Panel a-d) clearly demonstrates membrane localization of JIK in LNCaP cells. Panel e-h) shows no specific staining in DU145 cells which is JIK negative, demonstrating specificity. The images were captured at 60X magnification.

Target data

Serine/threonine-protein kinase that acts as a regulator of the p38/MAPK14 stress-activated MAPK cascade and of the MAPK8/JNK cascade. Acts as an activator of the p38/MAPK14 stress-activated MAPK cascade. In response to DNA damage, involved in the G2/M transition DNA damage checkpoint by activating the p38/MAPK14 stress-activated MAPK cascade, probably by mediating phosphorylation of upstream MAP2K3 and MAP2K6 kinases. Inhibits basal activity of MAPK8/JNK cascade and diminishes its activation in response epidermal growth factor (EGF).
