Supplementary information

The kinetochore is a protein complex that plays an important role during cell division particularly in ensuring proper chromosome segregation. It attaches to the centromere of each chromosome and serves as the anchor point for spindle microtubules. This interaction is essential for the movement and alignment of chromosomes on the metaphase plate. Although its exact molecular mass can vary due to its complex nature kinetochore is typically composed of multiple proteins including CENP proteins. It is found within the eukaryotic cells specifically expressed during mitosis and meiosis.
Biological function summary

Kinetochore is integral in forming a protein structure known as the kinetochore complex which is also called the spindle assembly checkpoint complex. It ensures chromosomes are properly aligned before the cell progresses to anaphase. Kinetochore also signals to delay anaphase until all chromosomes are correctly bi-oriented. Within this complex kinetochore proteins work together with regulatory proteins to prevent chromosome missegregation.

Pathways

Kinetochore plays an essential role in cell cycle regulation and mitotic spindle checkpoint pathways. It interacts closely with Aurora B and MAD2 proteins which both regulate the progression through mitosis. The mitotic spindle checkpoint pathway ensures chromosomes do not prematurely separate therefore maintaining genomic stability.

Kinetochore has a connection to conditions related to aneuploidy and cancer. Defective kinetochore function can lead to chromosome missegregation contributing to aneuploidy a feature observed in various cancers. Furthermore kinetochore malfunction is linked to Alzheimer's disease through its interaction with the tau protein which plays a role in neurodegenerative processes.

Target data

Essential component of the mitotic checkpoint, which prevents cells from prematurely exiting mitosis. Required for the assembly of the dynein-dynactin and MAD1-MAD2 complexes onto kinetochores (PubMed : 11146660, PubMed : 11590237, PubMed : 15824131). Its function related to the spindle assembly machinery is proposed to depend on its association in the mitotic RZZ complex.
