KLF2 also known as Kruppel-like factor 2 is a transcription factor with a molecular mass of approximately 37 kDa. It belongs to the Kruppel-like family of transcription factors which share zinc finger domains. KLF2 regulates gene expression by binding to specific DNA sequences in promoters influencing cellular processes. Researchers have localized its expression predominantly in the vascular endothelium the lungs and the spleen. In these tissues KLF2 influences endothelial cell function and vascular biology.
Biological function summary

KLF2 mediates cellular processes including endothelial cell maintenance and vascular development. As a transcription factor KLF2 does not operate as part of a multi-protein complex but rather performs its regulatory function individually by modulating target gene expression. KLF2 activates genes associated with anti-inflammatory and anti-thrombotic properties in endothelial cells contributing to vascular homeostasis. Its role is key in maintaining the balance between pro- and anti-inflammatory signals in the vascular system.

Pathways

KLF2 integrates its function into various signaling pathways notably the MEK/ERK and PI3K/Akt pathways. KLF2 modulates these pathways to control proliferation differentiation and apoptosis in endothelial cells. In particular KLF2-induced gene expression influences these pathways by interacting with other proteins such as MEK and ERK proteins that regulate cell survival and inflammatory responses. These interactions highlight the relevance of KLF2 in cellular signaling important for vascular health.

KLF2 has significant implications for cardiovascular diseases and atherosclerosis. Research associates KLF2 with protective roles against the development of atherosclerosis due to its influence on endothelial function and inflammation. When KLF2 expression is compromised the risk for thrombosis and vascular inflammation increases contributing to cardiovascular conditions. Additionally studies link KLF2 to the pathophysiology of diabetes where aberrant KLF2 expression disrupts normal endothelial functions exacerbating diabetic complications.
