KLF8 also known as Kruppel-like factor 8 is a transcription factor involved in regulating gene expression. It contains a zinc finger DNA-binding domain typical for the KLF family and weighs approximately 38 kDa. KLF8 expresses in several tissues with significant presence in kidney lung and liver. It shows strong nuclear localization important for its function in the control of transcriptional tone within cells.
Biological function summary

KLF8 regulates cell proliferation differentiation and development. This protein has been associated with a variety of cellular processes often functioning in cell cycle regulation by modulating the expression of genes involved in cell growth. KLF8 does not function as a part of a larger complex but interacts directly with DNA to exert its effects often affecting other KLF family members like KLF4 and KLF5.

Pathways

KLF8 plays an active role in the TGF-beta signaling and Wnt/beta-catenin pathways. It helps orchestrate cellular responses to growth factors by interacting with these pathways affecting processes such as epithelial-mesenchymal transition (EMT). In these pathways KLF8 associates with proteins such as beta-catenin and SMAD3 further emphasizing its role in cellular proliferation and differentiation.

KLF8 is known to relate to cancer and fibrosis. High KLF8 expression levels have been linked to the aggressiveness of certain cancers such as breast cancer and the protein's action influences EMT a critical step in metastasis. In fibrosis KLF8 contributes to pathological tissue remodeling. It connects with other proteins like MMP9 which are involved in matrix remodeling highlighting its significance in disease progression.
