KLHL12 also known as Kelch-like protein 12 functions mechanically as an E3 ubiquitin ligase. It belongs to the Kelch-like gene family and has a molecular mass of approximately 69 kDa. KLHL12 is expressed in various tissues particularly in the brain and neuronal cells where it plays significant roles in cellular processes. By facilitating protein degradation through ubiquitination KLHL12 regulates protein levels within the cell.
Biological function summary

This target is involved in the regulation of vesicle trafficking and activity through its interaction with various substrates. KLHL12 forms part of a cullin-RING E3 ligase complex which includes the CUL3 protein among others to mediate the ubiquitination of its target proteins. This process influences transport and secretion mechanisms having a critical impact on cellular function and communication.

Pathways

KLHL12 participates in the Wnt signaling pathway and the regulation of endoplasmic reticulum (ER) to Golgi transport. It plays a regulatory role in promoting the secretion of proteins such as collagen by facilitating vesicle formation. KLHL12 interacts with proteins like SEC31 and helps control protein trafficking within the secretory pathway which is essential for maintaining cellular homeostasis and response to external signals.

KLHL12 has links to neurodegenerative diseases and cancer. Abnormal KLHL12 activity or expression can contribute to certain neurodegenerative conditions by impacting neuronal protein homeostasis often associated with protein aggregates. In some cancers either KLHL12 or CUL3 levels are dysregulated potentially driving tumorigenesis by altering cellular growth pathways and protein degradation processes.

Substrate-specific adapter of a BCR (BTB-CUL3-RBX1) E3 ubiquitin ligase complex that acts as a negative regulator of Wnt signaling pathway and ER-Golgi transport (PubMed : 22358839, PubMed : 27565346). The BCR(KLHL12) complex is involved in ER-Golgi transport by regulating the size of COPII coats, thereby playing a key role in collagen export, which is required for embryonic stem (ES) cells division : BCR(KLHL12) acts by mediating monoubiquitination of SEC31 (SEC31A or SEC31B) (PubMed : 22358839, PubMed : 27565346). The BCR(KLHL12) complex is also involved in neural crest specification : in response to cytosolic calcium increase, interacts with the heterodimer formed with PEF1 and PDCD6/ALG-2, leading to bridge together the BCR(KLHL12) complex and SEC31 (SEC31A or SEC31B), promoting monoubiquitination of SEC31 and subsequent collagen export (PubMed : 27716508). As part of the BCR(KLHL12) complex, also acts as a negative regulator of the Wnt signaling pathway by mediating ubiquitination and subsequent proteolysis of DVL3 (PubMed : 16547521). The BCR(KLHL12) complex also mediates polyubiquitination of DRD4 and PEF1, without leading to degradation of these proteins (PubMed : 18303015, PubMed : 20100572, PubMed : 27716508).
