Supplementary information

KLHL21 also known as Kelch-like protein 21 is a part of the BTB-Kelch protein family. It is approximately 64 kDa in mass and plays a role in protein-protein interactions. KLHL21 acts as an adaptor that recruits specific substrates for ubiquitination. This protein has high expression levels in testis and moderate expression in various other tissues including the heart and kidney. Varying expression levels suggest a functional versatility across different cellular environments.
Biological function summary

KLHL21 forms part of the Cul3-based E3 ubiquitin ligase complex. This complex targets proteins for ubiquitination which marks them for degradation via the proteasome pathway. By mediating targeted protein turnover KLHL21 maintains cellular protein homeostasis. This is critical for regulating cell cycle progression and apoptosis highlighting its functional importance in maintaining cell integrity and function.

Pathways

KLHL21 integrates into cellular mechanisms through the ubiquitin-proteasome pathway. Its activity impacts protein degradation an important component of cellular regulation and signaling. KLHL21 interacts with important proteins like CUL3 part of the ubiquitin ligase complex assisting in the degradation of misfolded or damaged proteins. Proper function of this pathway is important for responding to cellular stress and maintaining cellular health.

KLHL21 links to various cancers due to its role in cell cycle regulation and apoptosis. The improper function or expression of KLHL21 can contribute to uncontrolled cell proliferation and tumor development. Additionally its interaction with CUL3 implicates KLHL21 in disorders like neurodegenerative diseases where ubiquitination and protein turnover are disrupted. Understanding KLHL21's role in these processes could aid in developing therapeutic approaches targeting these pathways.

Target data

Substrate-specific adapter of a BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex required for efficient chromosome alignment and cytokinesis. The BCR(KLHL21) E3 ubiquitin ligase complex regulates localization of the chromosomal passenger complex (CPC) from chromosomes to the spindle midzone in anaphase and mediates the ubiquitination of AURKB. Ubiquitination of AURKB by BCR(KLHL21) E3 ubiquitin ligase complex may not lead to its degradation by the proteasome.
