KLHL6 also known as Kelch-like protein 6 is a member of the Kelch-like family of proteins. It plays a mechanistic role in protein-protein interactions mainly by acting as a substrate recognition component of an E3 ubiquitin ligase complex. KLHL6 has a molecular mass of approximately 52 kDa. This protein is primarily expressed in B cells and it is most abundant in the germinal center B cells within the lymphoid tissues.
KLHL6 contributes to the regulation of B cell maturation and differentiation. Research indicates that KLHL6 is part of a Cul3-based E3 ubiquitin ligase complex which involves protein degradation processes. By functioning in this complex KLHL6 interacts with other proteins to ensure proper B cell development and immune responses. This function is important for maintaining normal immune system activities.
KLHL6 plays a significant role in the B cell receptor (BCR) signaling pathway. This pathway is essential for B cell activation and differentiation. KLHL6 is associated with other pathway-related proteins such as BCR components and signaling molecules downstream in the pathway. Another key biological pathway involving KLHL6 is the ubiquitin-proteasome system which regulates protein turnover and influences various cellular processes by targeting proteins for degradation.
KLHL6 alteration has been linked to certain B cell malignancies such as chronic lymphocytic leukemia (CLL). Its expression levels or mutations might influence the progression of these diseases. The protein is also connected with other ubiquitin-proteasome pathway components such as the Cul3 E3 ligase complex which might affect these disorders' development. Understanding KLHL6's role in these contexts can provide insights into potential therapeutic targets.
