In Western blot, ab239350 was shown to bind specifically to KMT3A / HYPB / HIF-1. A band was observed at 120-280, 440 kDa in wild-type HeLa cell lysates with no signal observed at this size in SETD2 knockout cell line ab261792 (knockout cell lysate ab257273). To generate this image, wild-type and SETD2 knockout HeLa cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 5 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times before development with Optiblot (ECL reagent ab133456) and imaged with 20 minutes exposure time. Secondary antibodies used were HRP conjugated Goat anti-Rabbit (H+L) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

KMT3A also known as HYPB (Huntington's disease gene-related protein B) or HIF-1 is a histone methyltransferase enzyme that mechanically transfers methyl groups to histone H3 at lysine 4 and lysine 36. This activity important for chromatin modification influences transcriptional regulation. The molecular mass of KMT3A is approximately 130 kDa. This enzyme is expressed in various tissues such as the brain heart and skeletal muscle highlighting its extensive physiological role.
Biological function summary

KMT3A modifies chromatin structure to regulate gene expression. Its histone methylation function plays a critical role in the modulation of transcriptional activity ensuring that genes are turned on or off when necessary for cellular function. Additionally KMT3A forms part of complex protein networks that mediate gene expression changes in response to cellular signals. Histone methyltransferase that specifically trimethylates 'Lys-36' of histone H3 (H3K36me3) using dimethylated 'Lys-36' (H3K36me2) as substrate (PubMed : 16118227, PubMed : 19141475, PubMed : 21526191, PubMed : 21792193, PubMed : 23043551, PubMed : 27474439). It is capable of trimethylating unmethylated H3K36 (H3K36me0) in vitro (PubMed : 19332550). Represents the main enzyme generating H3K36me3, a specific tag for epigenetic transcriptional activation (By similarity). Plays a role in chromatin structure modulation during elongation by coordinating recruitment of the FACT complex and by interacting with hyperphosphorylated POLR2A (PubMed : 23325844). Acts as a key regulator of DNA mismatch repair in G1 and early S phase by generating H3K36me3, a mark required to recruit MSH6 subunit of the MutS alpha complex : early recruitment of the MutS alpha complex to chromatin to be replicated allows a quick identification of mismatch DNA to initiate the mismatch repair reaction (PubMed : 23622243).
